چکیده
Glycosyl hydrolases are important mediators of plant cell wall modi®cation during plant development. These enzymes catalyse the hydrolytic release of speci®c sugars, such as L-arabinose, from the polysaccharide-rich cell wall matrix. The cloning and expression analysis of two genes, AtASD1 and AtASD2, encoding putative a-L-arabinofuranosidases in Arabidopsis thaliana are reported here. AtASD1 and AtASD2 identities were assigned on the basis of homology to plant and microbial family 51 glycoside hydrolases. Using RT-PCR, RNA gel blot analysis and reporter gene expression analysis, AtASD1 and AtASD2 were shown to have different developmental expression pro®les. High levels of AtASD1 promoter activity are present in multiple tissues during vegetative and reproductive growth. AtASD1 expression is particularly intense in zones of cell proliferation, the vascular system, developing and regressing ̄oral tissues, and ̄oral abscission zones. By comparison, AtASD2 expression is limited to the vasculature of older root tissue and to some ̄oral organs and ̄oral

Identification and Expression Analysis of Two Arabidopsis LRR-Protein Encoding Genes Responsive to Some Abiotic Stresses
AbstractTwo Arabidopsis thaliana genes, psr9.2 and psr9.4 appearedto be highly similar to a phosphate-starved induced gene,psr9, isolated from Brassica nigra suspension cells.Sequence analysis classified the encoded polypeptides asmembers of leucine-rich repeat (LRR) proteins superfamily. The sequence of psr9 proteins comprise a unique N-terminalregion e...
متن کاملDifferential Expression of Arabidopsis thaliana Acid Phosphatases in Response to Abiotic Stresses
The objective of this research is to identify Arabidopsis thaliana genes encoding acid phosphatases induced by phosphate starvation. Multiple alignments of eukaryotic acid phosphatase amino acid sequences led to the classification of these proteins into four groups including purple acid phosphatases (PAPs). Specific primers were degenerated and designed based on conserved sequences of PAPs isol...
متن کاملStructure and differential expression of the four members of the Arabidopsis thaliana ferritin gene family.
Four ferritin genes are found within the complete sequence of the Arabidopsis thaliana genome. All of them are expressed and their corresponding cDNA species have been cloned. The polypeptide sequences deduced from these four genes confirm all the properties of the ferritin subunits described so far, non-exhaustively, from various plant species. All are predicted to be targeted to the plastids,...

Background: TRR14 protein is a small member of a multi-gene family in Arabidopsis and is the first ones found during screening of seedlings for their resistant to the trehalose sugar. Objectives: Characterization ofTRR14 over-expressed plants with respect to morphological changes, growth and photosynthesis related parameters.Materials and methods: TRR14gene was isolated from Arabidop...
متن کاملAnalysis of SFL1 and SFL2 Promoter Region in Arabidipsis thaliana using Gateway Cloning System
SFL1 and SFl2 (SETH Four Like) genes are two members of SETH4 gene family in Arabidopsis thaliana expressed in saprophytic tissues. In this study, expression of SFL1 and SFL2 genes were studied using Gateway Cloning Technology. Primers were designed for PCR amplification of promoter region of SFL1 (900 bp) and SFL2 (930 bp) genes having attB1 recombination sites using Kod Hi Fi DNA polymerase e...
